SCANNING SERVICES
Microarrays are
scanned by Resource staff using one of three types of scanners,
dependant upon the labeling approach used in each series of
experiments. The
Axon GenePix 4000A
laser confocal scanner is used routinely for standard
amino-allyl labeling
and
Genisphere dendrimer
fluorescence at Cy3 and Cy5 wavelengths. Microarrays using
fluorophores outside of these two wavelengths are scanned on a
PE ScanArray 5000XL laser confocal scanner with 16
fluorophore capability including external blue laser excitation and
autoloader capacity.
Genicon RLS experiments using gold and silver particles
are scanned using a white light
HiLight CCD-based scanner for dual-wavelength scattered
light capture. All .tiff images are 16 bit and can be analyzed using
most academic freeware (e.g. Scanalyze) and commercially available
analysis packages (e.g. Axon GenePix Pro, Quantarray,
ArrayVision
etc).
Yale investigators may drop-off slides
for scanning at the Resource in Rm 2117
between 9:00 am and 4:30 pm Monday-Friday.
Slides should be brought in either a black slide box or a slide mailer
tightly covered with aluminum foil. Slides should
not be left exposed to fluorescent lab light.
Please complete a submission sheet, available when you
arrive or from the link below.
Yale Scanning Submission sheet
Image files are normally posted to Yale
Microarray Database on the same day they are dropped off via User ID
and password protected accounts. The accounts are based on PI or Lab
with each individual within a lab having his/her own ID and password.
Your User ID and initial password will be e-mailed to you after your
first scanning service. Please be sure you
have your own User ID and that it is entered on the
submission sheet for ALL subsequent
scans. This will ensure that the .tiff images are distributed to the
correct investigator.
Non-Yale investigators may print, complete
and FAX the Scanning Submission sheet at the link below. Please be sure
to complete all PI and billing information. A User ID and initial
password for YMD "Simple Menu" distribution of .tiff images will be
sent to you by e-mail. Please use the User ID on
ALL subsequent scan submissions.
Non-Yale Scanning Submission
sheet
Slides should be sent in carefully packed
slide mailers (see above) via overnight mail (Keck not responsible for
slide breakages or loss) to:
Attn: Lesa
Moemeka, Microarray Resource,
W.M. Keck Biotechnology Resource Lab at Yale, 300 George Street, Rm
2117, New Haven, CT 06511. Telephone (203) 785-5209.
Enter any special scanning needs, such as
fixed PMT values or non-standard fluorophore requirements, in the box
provided on the submission sheet.
Please contact Irina Tikhonova or
Lesa Clarke regarding
scanning questions.
