microarrays@yale.edu
Currently we have two Genomic Solutions-Gene Machines Omnigrid robotic arrayers with Telechem Stealth split pins housed in state-of-the art environmentally controlled Class100 cleanrooms. The Microarray Resource is able to work closely with individual investigators, Yale and Non-Yale, on a cost-recovery basis, to fabricate custom arrays. The Resource staff provide guidance and suggestions on array design based on their 8 years of experience.
Standard procedure for Custom Microarray
Contact Resource Associate Director (irina.tikhonova@yale.edu) via e-mail with brief outline of project.
Schedule a meeting to discuss requirements and time frame in detail.
Issue formal quotation for project.
Acceptance and implementation.
(The Resource reserves the ability to add additional charges, after discussions, if the project extends beyond the confines of the original scope of work).
In general the current rough estimate for each slide (minimum 100 slide per run) is approximately $85. All test prints, robotic transfers, plasmid preps, PCR, gel characterization, non-standard slides, print plates are recovered as separate supplemental charges outlined in the initial quotation.
Please contact Irina Tikhonova via e-mail or call (203) 785-5209 with questions.
Yale Keck Microarray Resource provides services on commertial and custom made spotted arrays which include labeling of RNA/DNA, hybridization, scanning and data analysis.
Labeling and Hybridization
Labeling and hybridization of RNA samples for gene expression analysis are done by using Genisphere Dendrimer technology.
Hybridizations of the arrays performed on ether Advalytix SlideBooster Autohyb Instrument or BioMicro MAUI hybridization stations. The ArrayBooster works on the principle of Surface Accoustic Wave technology to actively mix the liquid evenly over the array. The MAUI Hybridization Systems are also designed for highly reproducible hybridization conditions with controlled temperature and active mixing to provide high sensitivity and specificity hybridization results.
Scanning of the arrays performed on Axon GenePix 4200 laser confocal scanner. All .tiff images are 16 bit and can be analyzed using academic software (e.g. Scanalyze) and commercially available analysis packages (e.g. Axon GenePix Pro, Quantarray, ArrayVision etc).
Non-Yale investigators may print, complete and FAX the Scanning Submission sheet at the link below. Please be sure to complete all PI and billing information. A User ID for YMD "Simple Menu" distribution of .tiff images will be sent to you by e-mail. Please use the User ID on ALL subsequent scan submissions.
Non-Yale Scanning Submission sheet
Slides should be sent in carefully packed slide mailers via overnight mail (Keck not responsible for slide breakages or loss) to:
Attn: Irina Tikhonova , Keck Microarray Resource at Yale, 300 George Street, Rm 2110, New Haven, CT 06511. Telephone (203) 785-5209.
Enter any special scanning needs, such as fixed PMT values or non-standard fluorophore requirements, in the box provided on the submission sheet.
Statistical analysis of data is provided by the Keck Biostatistics Resource employing commercial softwares such as GeneSpring and a full range of academic softwares, algorithms and approaches on a cost recovery basis. Please contact Amy Lin for more details.
