Keck Microarray Core provides miRNA profiling services on Exiqon miRCURY LNA arrays.
microRNA expression profiling
miRCURY™ LNA Array microarray slides incorporate Exiqon’s unique Locked Nucleic Acid (LNA™) technology to provide high sensitivity and excellent mismatch discrimination for short microRNA (miRNA) targets. miRCURY™ LNA Arrays work on less than 1 µg total RNA (no need for microRNA enrichment) labeled using the miRCURY™ Array labeling kit and with a sensitivity of 50 attomol.
LNA™ Technology
Locked nucleic acid (LNA™) nucleosides are a class of nucleic acid analogues in which the ribose ring is “locked” by a methylene bridge connecting the 2’-O atom with the 4’-C atom. LNA™ nucleosides containing the six common nucleobases (T, C, G, A, U and mC) that appear in DNA and RNA are able to form base pairs with their complementary nucleosides according to the standard Watson-Crick base pairing rules.
However, by “locking” the molecule with the methylene bridge the LNA™ is constrained in the ideal conformation for Watson-Crick binding. LNA™ therefore makes the pairing with a complementary nucleotide strand more rapid and increases the stability of the resulting duplex. LNA™ nucleosides give rise to increased thermal stability and discriminative power of duplexes resulting in various unique features.
