MODIFIER PRICING
There are three factors in determining the cost of a modified oligo:
The Synthesis Cycle to be Used:
Determining the cost of a modified oligo begins
with selecting the scale of the synthesis and then calculating the setup charge
and the per- residue charge for that scale. A normal DNA setup charge is used
unless there is an RNA or 2'-O-methyl RNA residue at the 3' end for which those
setup charges would apply. A normal DNA per-residue charge is used for the
modified residues unless those modifiers are RNA or 2'-O- methyl RNA. Please see

Most of our specialy
reagents are ordered from
Glen Research. Glen offers only 0.2
micromole and 1.0 micromole columns for initiating syntheses with 3' modified residues (40 nanomole columns are not available). Therefore, the Normal 0.2 µm and Normal 1.0 µm
synthesis cycles are used for all syntheses with modified 3' residues.
The LV (low volume) 40 nm, LV 0.2 µm, or Normal 1.0 µm cycles are used for all
syntheses with unmodified 3' DNA residues. The LV cycles use approximately 70%
less reagent than the Normal 0.2 µm synthesis cycle.
The Wide Pore 1.0 µm cycle is used for all 1.0 µm oligos
over 50 bases in length.
Phosphorothioate linkages will be charged at $5
per linkage ($5.83 for non-Yale orders) to cover additional labor in setting up
and maintaining the synthesizer.
The Number of Additions per Bottle:
Glen generally offers specialty reagents in 50 micromole, 100 micromole, and 0.25 gram
bottles.
The number of additions available per bottle varies with each cycle and is displayed

The Prices of Specialty Reagents:
In addition to the set-up and per-residue
charges, the Glen Research
list price (+15% for non-Yale orders) for the needed amount of specialty
amidites and/or columns along with any ancillary reagents will be applied. For
specialty amidites from commercial companies other than Glen Research, that
company's list price +25% (+15% for non-Yale orders) will be applied. To
eliminate issues of stability of the unincorporated monomers, we special-order
the reagent for your oligos and discard any leftovers. We will work with you to
group your orders so as to minimize your cost by maximizing the utilization of
expensive monomers. If you prefer, we will work our way down a list of oligos
until a specialty reagent is exhausted.
Generally, we will incorporate ANY commercially
available modifier, not just those listed below, as long as it is
particulate-free. To avoid issues of improper storage and handling,
we recommend allowing us to order the modified phosphoramidite/column. However,
if you would like to provide us with a phosphoramidite/column that you purchased
or synthesized, a $250 surcharge for each different modification will apply.
This surcharge covers the additional labor costs of modified phosphoramidite/column
handling and storage, synthesizer programming, oligo processing and e-mail
correspondence. Also, since we are not supplying the modified phosphoramidite/column,
our usual quality guarantee does not apply and the oligos must be accepted "as
is". The following is a list of
commonly ordered specialty reagents and columns. Remember, we will incorporate
ANY commercially available modifier, not just those listed below, as long as it
is particulate-free.
