Protein HPLC: Preparative/With Fraction Collection
Intact proteins are fractionated on the Beckman-Coulter ProteomeLab PF2D system which uses chromatofocusing as the 1st dimension(pI=8.5 to 4.5) followed by non-porous reverse phase as the 2nd. An appropriate sample buffer is 7.5M urea, 2.5M Thiourea, 50 mM Tris, 6.25mM TCEP, and 1.25mM protease inhibitor and up to 1mL can be injected. The recommended minimum amount is 200μg.
